How does one go about determining if the floor of the bay is going to be strong enough? If it's laying flat on the floor, it has a giant foot print, so most likely not overly heavy in any one spot. I think making sure it is secure from shifting would be the bigger issue. It would be nice to just toss it in there and not worry about it, but that isn't the best option.

Bed frames are designed to be still all the time (well most of the time) the metallurgy of the bed frame may not work with all the jostling a bus does, brinelling issues and so forth. Just a thought

Two questions: Does it matter which side my black tank discharge valve is on and how does one start a new thread?

I have a mini skoolie and it would be easier to have the dump valve on the passenger side of the bus at this point.

Standard bed frames are made of spring steel, which is difficult to permanently "brinelling" or bend.

I hung mine similar to yours, but mine was only a thirty gallon tank. I hung one end using carriage bolts, with a plywood board centered and the other end was shimmed off a cross frame. Very solid. I then cradled the tank on three sides with two by fours, and the fourth side I injected foam between the tank and rear cross frame. It's solid as can be. We were t boned a few weeks back and that system didn't budge at all.

Yeah, water is heavy. I would put all thread up through the storage bay and catch the frame. Maybe consider every foot front and back of the tank and bridge it with unistrut
