Before any trip of any length I generally check the following.
Coolant level
Oil level and condition (the ol' sniff test just in case)
Tranny fluid level and condition
Power steering fluid (it leaks)
Air brake adjustment and lining thickness
Belt condition and tension
All lights
Tire pressure
Emergency supplies (fluids, tools, etc)
Battery charge level, connection condition, electrolyte level
General walkaround checking for leaks, missing stuff, loose stuff, etc
As for maintenance...I check my tire pressure, I adjust my brakes, I've changed the oil a few times, I've done fuel filters a few times, I dropped the pan on the tranny and did a filter and fluid, I drained and flushed the diff, I've greased stuff, and I've replaced some burnt out bulbs. My bus has been pretty hassle free and I think most of the stuff I've done was just preventive maintenance, but it's a good idea to check all of that. I'm sure you will find a routine and follow that. Actually, the CDL manual usually has a pretty darn good pretrip checklist you can go over.
