Smile Floor Holes

I'm about to finish my floor and need to seal the holes from the plywood that used to be there. I'm going to use Henry 212 unless I hear a strong reason to use something else. Should I seal them before I paint with Rustoleum or after? Not sure if the 212 is paintable or if that is advisable??

I painted first, then used aluminum tape to cover the holes.
